Identifying broad spectrum stress-responsive genes in Trametes versicolor to engineer enhanced tolerance to hydrothermal liquefaction aqueous phase
Listed in NCBI GEO
Industrial wastewater contains a variety of persistent pollutants that are difficult to treat and pose risks to ecosystems.
Description
One such stream, hydrothermal liquefaction aqueous phase (HTL-AP), is a toxic yet nutrient- rich byproduct generated during conversion of biomass to biocrude oil. HTL-APs are chemically diverse, posing significant challenges to implementing robust treatment methods for detoxification and nutrient reclamation.
Fungi exhibit a wide breadth of metabolic capabilities that respond dynamically to their environment, making them a resilient and versatile solution for degrading or transforming recalcitrant compounds from streams such as HTL-AP. Understanding the genetic basis of fungal tolerance in such environments is critical to fully realize their bioremediation capacity. This work leverages transcriptomic analyses to identify key gene families broadly associated with fungal stress tolerance, providing insight into the molecular mechanisms driving acclimation to various inhibitors.

The white-rot fungus Trametes versicolor was subjected to three distinct HTL-APs and identify consistently upregulated genes encoding NAD(P)-binding proteins, alpha/beta-hydrolases, aldo/keto reductases, and cytochrome P450s across all conditions, suggesting broad activation of redox and detoxification pathways. These results advance our understanding of fungal stress responses and lay the groundwork for future functional annotation efforts to support the development of new, stress-tolerant strains capable of reclaiming nutrients from diverse and hostile waste environments.
