Josef Schnuriger: Total synthesis of HMPQ and analogues for the visualisation of G-quadruplexes

To interrogate G4’s, prior studies have relied on the use of the BG4 antibody, which lacks DNA/RNA selectivity and is unable to be used on live cells significantly hindering the use of BG4.

The small molecule natural product HMPQ, has been shown to selectively bind to G4’s resulting in fluorescence. This property of HMPQ allows for selective visualisation of G4’s while its lack of toxicity suggests its potential for use in living cells. Developing synthetic methodology towards HMPQ allows the synthesis of structurally similar compounds which can fine tune the fluorescent properties and binding ability to improve HMPQ’s use as a G4 probe.</p>
