Effect of eIF5A K49 acetylation status on the translatome of Toxoplasma gondii

Listed in NCBI GEO

This study aims to investigate how the acetylation status of lysine 49 (K49) in eukaryotic translation initiation factor 5A (eIF5A) influences the ribosome translation landscape of Toxoplasma gondii.

Description

Using the ME49 strain as the genetic background, we performed ribosome profiling (Ribo-seq) on three distinct variants: wild-type parasites, an acetylation-mimic mutant (eIF5A-K49Q), and a deacetylation-mimic mutant (eIF5A-K49R).

Parasites were cultured under either standard conditions (non-stressed tachyzoites) or alkaline stress conditions (pH 8.2 medium for 4 days) to induce in vitro bradyzoite differentiation. Three biological replicates were included for each group, resulting in a total of 18 samples. By sequencing ribosome-protected mRNA fragments, this dataset enables genome-wide characterization of ribosome occupancy and translational output, providing a valuable resource for understanding how eIF5A-K49 acetylation reshapes translational programs during developmental stage conversion in Toxoplasma gondii.
