Research Data on the Mechanism of Hydroxysafflor Yellow A Regulating Autophagy to Ameliorate Renal Injury in Diabetic Mice
Listed in ScienceDB
This dataset originates from a comprehensive study based on network pharmacology and animal experiments to investigate the targets and mechanisms of Hydroxysafflor Yellow A (HSYA) in the treatment of Diabetic Kidney Disease (DKD).
Description
It consists of two parts: network pharmacology prediction and animal experimental validation. In the network pharmacology part, potential targets of HSYA (851) and DKD disease targets (4,905) were obtained from public databases, and their intersection yielded 402 key targets.
A protein-protein interaction network was constructed using the STRING database and Cytoscape 3.9.0, identifying core targets such as TP53, AKT1, and EP300. GO function and KEGG pathway enrichment analyses were performed using the DAVID platform, resulting in 764 BP terms, 104 CC terms, 195 MF terms, and 177 pathways (P < 0.05). The animal experiment part used a db/db mouse model to evaluate the protective effect of HSYA on DKD renal tissue injury.

The experiment was conducted in an SPF-grade animal experiment center from 2023 to 2025.The experimental subjects were 8-week-old male db/db mice and db/m mice, divided into 6 groups: db/m normal control group, db/db model group, valsartan group (10.4 mg/kg), HSYA high-dose group (10 mg/kg), HSYA medium-dose group (5 mg/kg), and HSYA low-dose group (2.5 mg/kg), with 5 mice in each group. All mice received continuous intragastric administration for 8 weeks.
This dataset contains individual observational records for 30 mice, with each record corresponding to the complete experimental data of one mouse. Row labels in the data table represent the numbers of individual animals. Column labels include: Group, Body weight (g), Blood glucose (mmol/L), Urinary albumin (mg/L), Urinary protein-to-creatinine ratio (mg/mmol), p-PI3K/PI3K protein expression ratio in kidney tissue, p-AKT/AKT protein expression ratio, and p-mTOR/mTOR protein expression ratio.
Western blot data were analyzed using ImageJ software for gray value analysis. Each protein sample was measured with 3 repeated detections, and the final results are presented as "mean ± standard deviation". All indicators involving protein ratios are dimensionless relative expression levels.
Urinary albumin and blood glucose detection were performed using commercially available kits according to the manufacturer's instructions, and the units of measurement are clearly indicated in the table. This dataset is submitted as a single Microsoft Excel file (.xlsx format). The file contains multiple worksheets: the first worksheet is the consolidated summary table of experimental data; subsequent worksheets contain embedded original Western blot images (each image corresponds to the development result of one protein target).
The Excel file can be opened normally in Microsoft Office 2019 or later versions, or in open-source software such as WPS Office and LibreOffice. The embedded images can be viewed directly within the worksheets. This dataset has no systematic missing data.
Data errors primarily originate from biological individual differences and operational factors such as antibody batch variations during Western blot experiments. The standard deviation of Western blot data generally ranges from 5% to 15% of the mean value, and the coefficient of variation for urinary albumin detection is controlled within 10%.
