F. Day 9 Dox-treated BCBL-1 iCas9 g<i>AAVS1</i> or <i>MARCHF5</i> g3 cells were treated with DMSO, 0.1 μM, or 0.25 μM of the MCL1 inhibitor S63845 for 2 days. Cell viability was measured with CellTiter-Glo 2.0 kit and expressed relative to gAAVS1. G. Reciprocal co-immunoprecipitation (co-IP) of the endogenous MARCHF5-NOXA complex in BC-3 cells.
Statistical differences were calculated using two-way ANOVA with Dunnett’s multiple comparison post-hoc test or unpaired t test as noted. Adjusted <i>p</i> values reflect post-hoc comparisons between each gRNA to gAAVS1 unless specifically noted. Error bars, standard error of the mean.</p>
