Polymorph control during continuous manufacturing

This project aimed to demonstrate a controlled process for polymorph conversion which could be operated on a continuous basis.
Description
Both monotropic and enantiotropic polymorphic systems were investigated, with the process required for the monotropic Forms I and II of Hydrochlorothiazide proving unsuitable for continuous manufacturing due to long hold times and a requirement for minimal agitation.<br><br>For the enantiotropic Forms III and I of Carbamazepine, a twin screw extrusion process was developed further and paired with quantification of of the polymorphic composition.
This system was able to operate at a steady state, while maintaining real-time data collection on the composition of the extruded powders.<br><br><i>Thesis is embargoed until 31 July 2027.</i>
