NGS of viral ds cDNA reads in plants

Listed in NCBI GEO

We previously demonstrated that two naturally occurring single-nucleotide mutations affecting distinct amino acids in the HCPro of a potyviral chimera independently increased virulence by enhancing either silencing suppression or proteolytic activity.

Description

To examine how beneficial variants arising during infection compete with the wild-type and with each other, we co-expressed pairs of these variants in Nicotiana benthamiana leaves.

We then evaluated their coexistence and interactions within host cells, as well as the impact of abiotic (elevated temperature) and biotic (silencing-impaired host) factors on infection dynamics. Outcomes were assessed in both local tissues at 5 days post-inoculation (dpi) and systemic tissues at 29 dpi.
