Single-Cell Analysis of an Intrinsic Reversal of Phototactic Behavior in Tilapia Larvae
Listed in NCBI GEO
Across vertebrates, parental care strategies are astonishingly diverse, yet they all provide specific environments that developing offspring must navigate in preparation for successful dispersal and independence while their brains continue to mature.
Description
Even though the transition from protected early life stages to independent living represents one of the most critical developmental milestones in animal life histories the neural underpinnings of offspring behavioral adaptations to these transitions remain poorly understood.
Here, we used single-nucleus RNA sequencing of whole larval brains of the Nile Tilapia, Oreochromis niloticus, a maternal mouthbrooder, to uncover the neural maturation processes associated with the dramatic developmental reversal in phototactic behavior during the transition from buccal incubation to freely swimming fry. We found that the behavioral switch from negative to positive light preference is accompanied by global brain maturation and a widespread remodeling of cell-type-specific gene co-expression networks, including a dramatic down-regulation of neural hemoglobin.

These results suggest a major shift in metabolic pathways as well as the homeostatic processes that regulate ion transport just as increasingly complex behavior and cognition emerge at the fry stage. When we mapped the single nucleus transcriptomes to the individual animals in the study, we discovered that expression of neural hemoglobin genes as well as mitochondrial and potassium-channel genes reflected the strength of individual behavioral responses.
Taken together, our study illuminates for the first time how metabolic and electrophysiological gene networks orchestrate the maturation of behaviorally relevant neural circuits during a key life-history transition.
