The UAP2-BRR2b Module Confers Cold Tolerance by Safeguarding Splicing Fidelity in Arabidopsis

Listed in NCBI GEO

Plants adapt to cold stress through transcriptomic reprogramming, yet how the core spliceosome maintains splicing fidelity under low temperature remains poorly understood.

Description

Here, we identify an U4/U6.U5 tri-snRNP-associated protein 2 (AtUAP2) as a cold-specific regulator of pre-mRNA splicing in Arabidopsis. Under normal conditions (22°C), the uap2, brr2b, and uap2 brr2b mutants exhibit no developmental anomalies.

Upon cold stress, however, uap2 displays severe and specific cold sensitivity. Transcriptome-wide analysis revealed that UAP2 deficiency leads to widespread mis-splicing under low temperature. Expressing correctly spliced AtGRP7 and AtLOS2 cDNAs in uap2 partially rescues its cold sensitivity, confirming both as downstream effectors.

Mechanistically, UAP2 physically interacts with the U5 RNA helicase BRR2b. While the brr2b mutant displays cold sensitivity similar to uap2, the uap2 brr2b double mutant exhibits an even more pronounced cold-sensitive phenotype. Crucially, this genetic synergy and cold-specific vulnerability demonstrate that the UAP2-BRR2b complex operates as a dedicated functional module required to sustain spliceosomal efficiency under cold stress, thereby providing a key mechanistic link between core splicing machinery dynamics and plant cold tolerance.
