Subcutaneous immunization with replication-competent live influenza virus confers durable protective immunity without respiratory pathology in mice
Listed in ScienceDB
Current influenza vaccination strategies are constrained by the trade-off between the safety of inactivated platforms and the immunogenic potency of live attenuated vaccines.
Description
Here, we provide a comprehensive demonstration that that subcutaneous (SC) immunization with a non-attenuated, replication-competent live influenza virus (LIV) elicits rapid, robust, and durable protective immunity while maintaining a favorable safety profile.
A single high-dose LIV SC immunization elicited strong neutralizing antibody responses and conferred complete protection against lethal homologous virus challenge, outperforming intramuscular (IM) delivery, inactivated virus, and recombinant hemagglutinin–based vaccines. Viral replication following SC immunization was transient and spatially restricted, with rapid clearance and no detectable lung infection or tissue pathology.

LIV SC immunization triggered early innate activation in draining lymph nodes, followed by robust germinal center (GC) responses, plasma cell differentiation, and memory B cell formation. Single-cell transcriptomic analysis revealed a temporal transition from interferon-driven antiviral programs to coordinated antigen presentation and humoral immune differentiation. Together, these findings establish LIV SC immunization as a non-respiratory strategy that preserves the immunological potency of live virus exposure while minimizing pathogenic risk, providing a conceptual framework for the development of rapid-response influenza vaccines.
