Mechanism of Sishen Pill on Ulcerative Colitis based on Network Pharmacology and animal experiments

A mouse model of UC with spleen and kidney yang deficiency was constructed, and Sishen Pill was given intervention treatment. The network pharmacological prediction was verified by WB and RT-qPCR experiments.Results The core components of Sishen Pill in the treatment of UC include quercetin, β-sitosterol, cholecalciferol, delphinium pigment, puerarin, etc. The core targets include IL-6, TP53, EGFR, AKT1, IL1β, etc., and mainly play a role through IL-17, PI3K-Akt, Toll-like receptor, TRP signaling pathway, etc.

The core component has good binding performance to the core target; Sishen Pill can reduce the expression of EGFR and IL-6 and increase the expression of p53 in mouse colon tissue.Conclusion Sishen Pill can regulate EGFR, IL-6, p53 and other targets, regulate IL-17, PI3K-Akt, Toll-like receptors, and TRP signaling pathways, reduce inflammatory damage and treat UC.
