Description

Abstract

This dataset comprises the raw experimental data supporting the core conclusions of the research article entitled "Highly Specific Pump-Probe Photoacoustic Tomography Based on Rigidity-Induced Long-Lived Triplet State Methylene Blue." It addresses the issues of endogenous thermal background interference in pump-probe photoacoustic imaging and the limited imaging specificity caused by the short triplet-state lifetime of conventional liquid-phase probes.

The dataset includes steady-state spectroscopic characterization data of methylene blue (MB) in four different microenvironments (aqueous solution and PVA solid films with three degrees of alcoholysis), transient triplet differential (TTD) signal decay kinetics, and raw signal data from pump-probe photoacoustic imaging background-suppression experiments. The data files cover all raw data corresponding to Figures 2–4 of the paper, in formats including .csv and .mat, which can be accessed using common software such as Excel, Origin, or MATLAB (R2016a and above).

The raw photoacoustic waveform data (approximately 6 GB for imaging experiments) are not included in this dataset due to their large size, but are available from the corresponding author (Wang Bo, Email: wangbo0302@csu.edu.cn) upon reasonable request. This dataset serves as the core experimental data for validating the rigidity confinement strategy for enhancing pump-probe photoacoustic imaging specificity, and can be reused by researchers in related fields.Figure 2(a) Steady-state absorption spectra (Data filename: 吸收谱对比)This dataset contains the raw steady-state UV-Vis absorption spectra of methylene blue (MB) in aqueous solution and PVA-1799 solid film.

Spectra were acquired using a N4S dual-beam UV-Vis spectrophotometer (INESA, Shanghai) over a range of 400–900 nm with a step size of 1 nm. The file is in two-column comma-separated values format: the first column is wavelength (nm), and the second column is absorbance (a.u.). The data are directly output from the instrument without any smoothing or normalization.

The normalized UV-Vis absorption spectra shown in Figure 2(a) were obtained by truncating the data to 400–800 nm and normalizing to the 0–1 range.Figure 2(b) Steady-state fluorescence emission spectra (Data filenames: MB-PVA荧光光谱, MB荧光光谱, each containing 12 scans)This dataset contains the raw steady-state fluorescence emission spectra of MB in aqueous solution and PVA-1799 solid film. Spectra were acquired using an Ocean Optics USB2000+ fiber-optic spectrometer with a 660 nm continuous-wave laser as the excitation source, scanning from 339.977 to 1026.136 nm, with an integration time of 0.5 s and dark noise correction enabled.

Each compressed folder contains 12 repeated measurements. The files are in two-column comma-separated values format: the first column is wavelength (nm), and the second column is the dark-noise-subtracted raw intensity (counts). The data are directly output from the spectrometer without any smoothing or normalization.

The normalized fluorescence emission spectra shown in Figure 2(b) were obtained by averaging all measurements, truncating the data to 670–900 nm, and normalizing to the 0–1 range.Figure 3 TTD lifetime data and photoacoustic data (Data filenames: MB_TTD_data.mat, MB_PV75_TTD_data.mat, MB_PVA88_TTD_data.mat, MB_PVA99_TTD_data.mat, TTD_data.mat)This dataset contains the transient triplet differential (TTD) signal decay data of MB molecules as a function of pump-probe delay time in four different microenvironments: aqueous solution (MB), low-alcoholysis PVA-75% film (MB_PVA75), PVA-1788 with 88% alcoholysis (MB_PVA88), and PVA-1799 with 99% alcoholysis (MB_PVA99).

The data are extracted results after photoacoustic signal differential processing. The TTD decay curves shown in Figure 3(a) and the triplet-state lifetime bar chart in Figure 3(b) were both fitted and plotted based on this dataset. TTD_data.mat contains the reconstructed photoacoustic data at nine delay times, corresponding to the TTD spatiotemporal evolution images shown in Figure 3(c).

All data files can be directly loaded and read using MATLAB (R2016a or later). The raw photoacoustic waveform data (approximately 3 GB) are not included in this dataset due to their large size, but are available from the corresponding author (Wang Bo, Email: wangbo0302@csu.edu.cn) upon reasonable request.Figure 4 Pump-probe photoacoustic imaging data (Data filenames: PVA_MB-Hb_delay=2.mat, TTD_data.mat, 固态PVA-MB薄膜与纯Hb样品管的归一化TTD信号.mat, Hb-MB混合液样品管与纯Hb样品管的归一化TTD信号.mat)This dataset contains the raw data from pump-probe photoacoustic imaging background-suppression experiments, corresponding to Figure 4 of the paper:Figure 4(a): Pump-probe photoacoustic imaging results of solid PVA-MB film versus pure Hb tube at a probe delay of 2 µs (including M-image, P-image, T-image, and TTD-image).

The raw photoacoustic waveform data (approximately 6 GB for imaging experiments) are not included in this dataset due to their large size, but are available from the corresponding author (Wang Bo, Email: wangbo0302@csu.edu.cn) upon reasonable request.
