Mechanism of Polygonatum cyrtonema Hua polysaccharide PCP1 in improving obesity by modulating calcium signaling pathways

This study investigate the effects and potential mechanisms of Polygonatum cyrtonema Hua polysaccharide 1 (PCP1) against obesity.

In vitro, cocktail-induced differentiation of 3T3-L1 preadipocytes was treated with PCP1 at 25, 50, 100 μg/mL and evaluated using Oil Red O staining, RT-qPCR, and pharmacological inhibition or activation of calcium signaling. PCP1 reduced lipid droplet accumulation in a concentration-dependent manner and downregulated the gene expression of peroxisome proliferator-activated receptor gamma (Pparg) and CCAAT/enhancer-binding protein alpha (Cebpa).
Experiments using 2-aminoethoxydiphenyl borate (2-APB) and ionomycin indicated that the inhibitory effect of PCP1 on adipocyte differentiation was at least partially dependent on calcium signaling. In vivo, ob/ob mice were assigned to model, semaglutide, and PCP1 groups, with wild-type mice serving as controls, and treated for 8 weeks. PCP1 reduced body weight and the weights of subcutaneous adipose tissue, epididymal adipose tissue, and liver; improved fasting hyperglycemia, blood lipids disorders, glucose intolerance, and insulin resistance; alleviated hepatic steatosis, lipid deposition, and subcutaneous adipocyte hypertrophy; and decreased alanine aminotransferase (ALT) and aspartate aminotransferase (AST) levels.

PCP1 also increased uncoupling protein 1 (UCP1) expression in brown adipose tissue, whereas no evident abnormalities were observed in blood urea nitrogen (BUN), creatinine (Cr), or the histopathological examination of the heart, spleen, lung, and kidney. Transcriptomic analysis identified 649 differentially expressed genes, mainly enriched in calcium signaling, fatty acid metabolism, fatty acid biosynthesis, and thermogenesis.
RT-qPCR confirmed that PCP1 downregulated calcium signaling-related genes, including inositol 1,4,5-trisphosphate receptor type 3 (Itpr3), voltage-dependent anion channel 3 (Vdac3), Fms-related receptor tyrosine kinase 1 (Flt1), and solute carrier family 8 member A1 (Slc8a1); upregulated genes involved in fatty acid β-oxidation and thermogenesis, including sirtuin 1 (Sirt1), Ucp1, peroxisome proliferator-activated receptor alpha (Ppara), and peroxisome proliferator-activated receptor gamma coactivator 1-alpha (Ppargc1a); and suppressed genes involved in lipid synthesis and uptake, including fatty acid synthase (Fasn), diacylglycerol O-acyltransferase 2 (Dgat2), stearoyl-CoA desaturase 1 (Scd1), and cluster of differentiation 36 (Cd36).
Collectively, PCP1 may ameliorate obesity and associated metabolic abnormalities by modulating calcium signaling, enhancing fatty acid β-oxidation and thermogenesis, and suppressing lipid synthesis, supporting its potential development as a functional food ingredient for obesity management.
