Integrative spatiotemporal transcriptomic and spatial immune repertoire sequencing of the JEV-infected mouse brain elucidate the antiviral immune response landscape
Listed in ScienceDB
Japanese encephalitis virus (JEV) spread wildly and caused thousands of deaths.
Description
Its infection mechanism is not clearly characterized and there are no approved drugs as well. Here, spatial transcriptomics and novel spatial immune repertoire sequencing of the mouse brain were utilized to reveal the dynamics of tissue composition, transcriptome, and immune repertoire during the JEV infection.
It was demonstrated that the blood-brain barrier (BBB) proportion increased and was associated with the immune cell infiltration and the JEV was preferred to infect and replicate in the cortex (CTX). It was implied that infiltration of monocytes, macrophages, and dendritic cells in the BBB was correlated with the early immune response. It’s revealed that T cell receptor (TCR) clonotypes less infiltrated and expanded in the fiber tract (FB) compared to the BBB and the CTX.

Hydrophilic TCR clonotypes infiltrated the brain and replicated in the FB post-infection. All boosted the study of the mouse brain of the JEV infection into the era of spatial immune repertoire and transcriptome indicating the immune response.
