A Kompetitive Allele Specific PCR (KASP) assay was developed and validated to detect the G143A single nucleotide polymorphism (SNP) in the cytochrome b gene (cytB) of Stemphylium vesicarium, associated with resistance to quinone outside inhibitor (QoI; FRAC Group 11) fungicides.
Description
The assay was applied to: (i) a collection of 70 single-spore isolates from onion fields in the Holland Marsh, Ontario from 1995 to 2023; (ii) DNA extracted directly from Burkard multi-vial cyclone sampler tubes collected at the Ontario Crops Research Centre - Bradford in 2021 and 2022; and (iii) single-spore isolates recovered from Burkard collections in 2023-2024.
The G143A mutation was detected in 67 of 70 historical isolates, and in all single-spore isolates recovered from Burkard collections in 2023-2024.
