COMPASS-like complex participates in the bud dormancy release of tree peony by regulating H3K4me3 modification
Listed in NCBI GEO
Bud dormancy release in woody plants is crucial for survival, regrowth, flowering and fruiting.
Description
Tree peony (Paeonia suffruticosa), an importantly ornamental and economic plant undergoes bud endodormancy in winter, and effective chilling duration and exogenous gibberellins (GA) can effectively break the dormancy, however, its epigenetic regulation mechanism remains poorly understood. Here, western blotting revealed that H3K4me3 but not H3K4me1 and H3K4me2 was associated with chilling- and gibberellin (GA3)- induced dormancy release.
Chromatin immunoprecipitation sequencing (ChIP-seq) combined with RNAseq results revealed that H3K4me3 enriched near transcription start sites (TSS), and was correlated with the transcriptional activation of dormancy release-related genes (PsEBB3, PsCYCD3.1, PsCYCD3.3, PsBG6, and so on). H3K4me3 enrichment genes (HEGs) common to the differentially-expressed genes (DEGs) were mainly enriched in KEGG pathways such as plant hormone signaling transduction and MAPK signaling.

Four COMPASS-like component homologs were screened, among them, PsWDR5a, PsRBL, and PsASH2R were up-regulated by the prolonged chillings and GA3 treatment. PsRBL interacted with PsWDR5a and PsASH2R using yeast two-hybrid, luciferase complementation. Yeast three- hybrid and co-immunoprecipitation assays confirmed that PsRBL bridged the other proteins to form a nuclear-localized complex.
Functional analyses showed that PsWDR5a, PsRBL, and PsASH2R significantly promoted budburst and elevated genomic H3K4me3 levels. Our fingings would provide novel insights into the epigenetic regulation of dormancy transitions in woody perennials.
