Phytosulfokine peptide regulates the growth-immunity trade-off through CAMTA3-mediated transcriptional signaling in Arabidopsis

Phytosulfokine (PSK) peptide hormone promotes plant growth while suppressing endogenous immunity.

However, the underlying mechanism remains poorly understood. Here we show that PSK regulates the trade-off between growth and immunity by mediating stabilization and cytosol-nucleus translocation of Calmodulin-binding Transcription Activator 3 (CAMTA3).
By genetic screening of an Arabidopsis thaliana mutant library, we identified a mutant named growth retardation and autoimmunity 1 (gra1). Map-based cloning and bulk population sequencing disclosed that GRA1 encodes TYROSYLPROTEIN SULFOTRANSFERASE (TPST) for the biosynthesis of sulfated peptides. Treatment with PSK can restore the reduced callus proliferation of gra1 mutants and overexpression of PSK can promote plant growth but reduce immune responses.

RNA-seq and genetic studies show that PSK regulates plant growth and immunity through salicylic acid pathway. Further, we revealed that PSK stabilizes CAMTA3 and enables its translocation from cytosol to nucleus by facilitating its binding to calmodulin 6 (CaM6) and CaM7, and CAMTA3 inhibits the expression of SYSTEMIC ACQUIRED RESISTANCE DEFICIENT 1 (SARD1) and CALMODULIN-BINDING PROTEIN 60g (CBP60g) to inhibit salicylic acid biosynthesis and plant immunity.
Collectively, we disclose a signaling pathway PSK-PSKR-CaM6/7-CAMTA3-SARD1/CBP60G that regulates the trade-off between plant growth and immunity.
