Effects of combined shading and mulching ecological practices on microclimate, soil properties and bacterial communities in tea plantations

Natural ecological measures (shading and covering) have a synergistic effect on the tea garden ecosystem, but their micro ecological driving mechanisms are not yet clear.

Description

This study focuses on the Fuding Dabai tea tree as the research object, and sets up four treatments: open field control (CK), shading between cherry blossom trees (S), pruning between branches and leaves (M), and shading coverage composite (SM). Microclimate temperature, humidity, and soil physicochemical properties are measured, and the structure, diversity, and functional characteristics of soil bacterial communities are analyzed.

The experiment was carried out in the tea garden (36 ° 22 ′ N, 116 ° 52 ′ E) of Mount Taishan Tea Valley Agricultural Development Co., Ltd. from August 2023 to August 2024. The temperature and humidity indicators of the microenvironment were measured using the Pengyun IoT temperature and humidity detector (model: S21A, accuracy ± 0.1 ℃, ± 1% RH) for three consecutive days from August 14-16, 2024.

Measurements are taken at 10:00 every day, with 3 measurement points set up in each community, and each point is repeated 3 times to take the average value. The measurement indicators include: temperature and humidity of the 10 cm soil layer, temperature and humidity of the tea tree canopy (20 cm above the canopy), and temperature and humidity of the ground (5 cm above the ground). The soil samples were collected using the five point sampling method in small areas on August 15, 2024.

Soil physical and chemical properties: pH value, organic matter (OM), alkaline nitrogen (AN), available phosphorus (EP), and available potassium (AK) content are determined according to the "Soil Agricultural Chemical Analysis Method". Soil samples were collected using FastDNA ® SPIN Kit (MP Biomedicines, USA) kit for extracting genomic DNA. Use specific primers with barcode to amplify the V3+V4 variable region of 16S rDNA.

The primer sequences used are: forward primer 341F: CTACGGGNGWGCAG and reverse primer 806R: GGACTACHVGGGTATCTAAT. After purification, the amplified product was connected to a sequencing adapter and sequenced using the Illumina Hiseq 2500 platform. After obtaining raw reads through sequencing, DADA2 software is used for data filtering and quality control to obtain high-quality clean data.

Based on the sequence and abundance data of amplification sequence variants (ASVs), subsequent analysis is carried out. The analysis of soil bacterial community structure, alpha diversity and beta diversity, differential species analysis, microbial community function analysis, and environmental factor correlation analysis were all conducted on the OmicSmart cloud platform( omicsmart.com )Completed on top.
