P29-20 Establishing realistic dry aerosol exposure conditions at the Air-Liquid Interface with an in vitro alveolar model [* Cross-Reference *]

Listed in LORE

The MACRAMÉ project (for Advanced Characterisation Methodologies to assess and predict the Health and Environmental Risks) aims to establish robust and harmonized methodologies for assessing the safety of inhalable nanomaterials (NMs) and advanced materials.

Description

Within this framework, the suitability of in vitro alveolar models combined with dry aerosol exposure systems is being explored to address current data gaps in the toxicological evaluation of nanomaterials under physiologically relevant conditions.

In this study, a monoculture of A549 cells grown on transwell inserts at the air-liquid interface (ALI) was used as an alveolar barrier model. To mimic inhalation more realistically, exposure was conducted using the Vitrocell® PowderX system, which enables the aerosolization of dry materials directly onto the cellular surface. Reference materials including Quartz DQ12, Corundum nanoparticles, multi-walled carbon nanotubes (NM-401 from JRC), and Graphene particles (NM48001a from JRC) were used.

Deposition of the aerosolized materials onto the cellular surface was quantified by two independent methods: gravimetric analysis and surface-sensitive quartz crystal microbalance (sQCM). Scanning electron microscopy (SEM) was used to qualitatively confirm particle localization. Cell viability was assessed using the Alamar Blue assay, while intracellular reactive oxygen species (ROS) production was measured via flow cytometry.

Pro-inflammatory cytokine release was analyzed in both apical and basolateral compartments using Luminex-based multiplexing. In addition, particle–cell interactions and internalization were visualized through light microscopy on Richardson-stained cell cross-sections. Even though the PowderX device allows only limited throughput, with just four wells exposed at a time, making it less suitable for large-scale screening, it enables accurate and reproducible dose-response curves for dry powder exposure, demonstrating its suitability for studies requiring realistic exposure scenarios without the complications of dispersion protocols. ***This entry has been automatically imported via BNL CSV by LIST harvest scripts.
