The succinylomics analysis in WT liver tissues and SIRT5 KO liver tissues of MASH model mice
Listed in Teesside University Research Data Repository
To explore the potential role of SIRT5 in MASLD, SIRT5 knockout (SIRT5 KO) mice were purchased from Cyagen Biosciences (Guangzhou, China) and housed in an SPF facility.
Description
Sirt5 KO mice and WT (C57BL/6J) mice were fed an high fat, high fructose and the addition of cholesterol (HFF) diet for 16 weeks. and succinylomics analysis was performed by Novogene Co., Ltd. (Beijing, China).
We conducted GO or KEGG enrichment analysis on the differentially expressed genes for each pairwise comparison using the EnrichPipeline workflow (Bioinformatics enrichment tools: paths toward the comprehensive functional analysis of large gene lists. Nucleic Acids Res 37: 1-13.). Proteins with Log2FC > 2.0 were included, and P < 0.05 was considered statistical significance.

R-4.5.3 and R Studio software is used to perform calculations and visualizations. Our research results indicate that the absence of SIRT5 accelerates the hepatic steatosis, inflammation and fibrosis in MASH. SIRT5 can remove succinyl-CoA modification from various key metabolic enzymes, thereby regulating the metabolic process and ultimately improving the condition of MASH.
