Structure-function studies of Yersinia spp. lipid A

Listed in ZivaHub and Deakin Research Online and DMU Figshare — shown once because both records carry DOI 10.17034/32638182.v1

The Yersinia genus includes human pathogens: Y. pestis, Y. pseudotuberculosis, and Y. enterocolitica, and 14 non-pathogenic species.

Description

The expression of virulence factors is temperature regulated in pathogenic yersiniae. In pathogenic Y. enterocolitica and Y. pestis at 21 °C, the lipid A is hexa-acylated and may be modified with aminoarabinose and/or palmitate.

However, at 37 °C, it is mainly tetra-acylated and related to immune-evasion. In the present work, we have expanded this knowledge by including the Y. enterocolitica pathogenic serotypes 0:3 and 0:9, which did present the same lipid A pattern at 21 °C and 37 °C as the 0:8 serotype. By combining genetic and mass spectrometry analysis, we demonstrated that lipid A from non-pathogenic Yersinia spp. is different from pathogenic species.<br><br>Tetra-acylated lipid A structures are not found in non-pathogenic Yersinia.

Moreover, we show that non-pathogenic Yersinia encode for a novel LpxL acyltransferase able to modify the lipid A with two or only one fatty acid. 3-D modelling and docking experiments showed that IpxL from Ye08 (LpxLoe) and Y. aldovae (LpxLy.awo^e) have minor genetic differences. In this work we demonstrate that these single difference is the responsible for the specificity of LpxLos for only one substrate instead of two.<br>Our data revealed that the expression of the two component systems PmrA-PmrB and PhoP-PhoQ are temperature-regulated in the entire genus, suggesting that the lipid A is also temperature regulated.<br><br>We generated a Y. enterocolitica 0:8 (Ye08) expressing a non-pathogenic lipid A, and we investigated the implications of this lipid A on the expression/function of Y. enterocolitica virulence factors.

We showed that motility is up-regulated, and we mechanistically established that the expression of flhDC, regulator controlling the flagellar regulon, is down- regulated. Additionally, we have shown that alteration in epithelial cells invasion by this strain is only caused by the absence of LpxR. In contrast, the levels of the virulence plasmid encoded virulence factors Yops and YadA are not affected.

Finally, we establish that Ye08 expressing non-pathogenic lipid A induce higher activation of the LPS receptor.
