The triterpenoid monomer from Butyriboletus roseoflavus ameliorates alcoholic liver disease related to Bifidobacterium animalis‑mediated suppression on hepatic Th17 cells
Listed in Teesside University Research Data Repository
Natural products targeting the gut-liver axis offer a promising source of therapeutic candidates for alcoholic liver disease (ALD), a condition with limited current treatment options.
Description
Here, Butyriboletus roseoflavus triterpenoid complex was prepared and column-purified, exerting prominent hepatoprotective effects against both acute and chronic ALD. Subsequent in vitro screening identified 16α‑Hydroxytrametenolic acid (HTA) as the principal active monomer for the hepatoprotective activity.
HTA treatment significantly alleviated alcohol‑induced hepatic steatosis, inflammation, and fibrosis, as evidenced by histopathological and biochemical analyses. Mechanistically, HTA markedly enriched intestinal Bifidobacterium animalis and upregulated the expression of phosphotransacetylase (Pta), a pivotal enzyme responsible for acetate biosynthesis. Both endogenous and exogenous acetate protected hepatocytes from alcohol-induced injury in vitro.

In vivo HTA suppressed IL‑6/STAT3/RORγt signaling and Th17 differentiation. Our findings establish HTA as a novel prebiotic-like agent that enriches B. animalis and drives acetate-ediated immune responses to ameliorate ALD. This study highlights the translational potential of integrating fungal metabolites with probiotic strategies.
