<p>A peptide vaccine containing bacterial epitopes of CD4<sup>+</sup> T cells is immunogenic in mice.</p>

<p>(A) C57Bl/6 mice were immunized with a peptide vaccine containing five peptide epitopes of <i>Francisella</i> (FTL_1498-KE-15, MinD-IR-16, FTL_1678-YK-16, DnaK-AE-16, DacD-RK-17), and the cellular response in the spleen and lymph nodes was assessed using the ELISpot assay.

A group of mice (control) received the same adjuvant formulations but without peptides. (B) Number of IFN-γ-producing cells in splenocytes following stimulation with vaccine-contained peptides.

Responses were determined in individual mice from the peptide vaccine group (PepVac, n = 6) or the control group (control, n = 3). Measurements were performed on days 13, 17 and 19 after the booster dose (due to capacity reasons) and the data are combined in this graph. Values for individual mice (points) and their averages are shown.

Note

The unequal group sizes were due to the limited availability of animals. (C-E) Number of cells producing IFN-γ, IL-2 and TNF-α in splenocytes following stimulation with a subset of peptides contained in the vaccine. Splenocytes were isolated 19 days after administration of the booster dose and pooled from groups of 3 mice.

Data from individual wells for each condition and the corresponding mean values are shown. For all assays, peptides were used at a final concentration of 5 µM, and control wells without peptide (unstim) or with a negative peptide (YAIAVNDLGTEYVHR) were included. Significant differences compared to the control (unstim) were identified using two-way ANOVA followed by Dunnett’s post hoc test.

Asterisks indicate p-values (*p < 0.05, **p < 0.01, ***p < 0.001, ****p < 0.0001). This experiment was performed once.</p>
