53BP1 condensate regulates the completion of heterochromatin DNA replication

The epigenetically silent heterochromatin is replicated at a specific window during the cell cycle; however, the mechanisms governing the completion of heterochromatin replication remain elusive.

Description

Here, through mass spectrometry to analyze factors enriched in P53-binding Protein 1 (53BP1) condensates, we report that 53BP1 is intricately involved in complexes governing both DNA replication and epigenetic mark restoration at heterochromatin.

Depletion of 53BP1 resulted in reduced recruitment of epigenetic factors at heterochromatin, impaired heterochromatin epigenetic mark restoration after DNA replication, and prolonged heterochromatin replication. These defects can be rescued by wild type or condensate proficient, but not deficient, 53BP1. Condensate formation and heterochromatin replication regulation of 53BP1 require its interaction with Proliferating Cell Nuclear Antigen (PCNA).

Further, the interaction with PCNA mediates the transition of 53BP1 from condensate to DNA damage repair. Together, our results support an important role of 53BP1 condensate in regulating the completion of heterochromatin replication.
