Spatial confinement promotes germinal center diversity and tempers immunodominance [ZIP-Seq]
Listed in NCBI GEO
Germinal centers (GCs), sites of antibody somatic mutation and B cell selection, form as confined clusters of motile cells within individual lymphoid follicles.
Description
Each responding lymph node (LN) can have multiple GCs, and GCs typically do not exchange cells with one another once seeded in the LN. While this isolation of GCs has long been appreciated in lymphoid tissue anatomy, the benefits or drawbacks of this actively maintained organization on the GC response have been unclear.
Here, using mice whose peripheral LNs exhibit GC dispersal due to B cell deficiency in Ga13-dependent G-protein-coupled receptor (GPCR) activity, we investigate the impacts of GC B cell de-confinement on the evolutionary events following immunization. We find that GC-to-GC trafficking can occur in confinement-defective LNs, and that fluorescent reporter heterogeneity between GCs in multi-color GC lineage tracing mice becomes lost.

When analyzing the GC B cell receptor (BCR) repertoire using spatial approaches, we observed more expansions of the same clone in opposite ends of a LN and more crossover between neighboring GCs within clonal lineages when GCs were dispersed. The total GC B cell repertoire of individual LNs also showed decreased overall diversity and an outsized contribution from a small number of clones in the de-confined condition. As an outcome, dispersed LNs favored clones responding to immunodominant over subdominant epitopes to a greater degree than normal LNs.
Together, our findings suggest that confinement supports antibody breadth by moderating the spread of dominant clones and preserving unique trajectories in individual GC responses.
