Transcriptomic profiling of vemurafenib-resistant versus parental BRAF V600E mouse melanoma cells (5555 and 5555R)

Listed in NCBI GEO

Resistance to BRAF inhibitors (BRAFi) limits long-term clinical benefit in BRAF-mutant melanoma.

Description

To characterise tumour-intrinsic transcriptional changes associated with acquired BRAFi resistance, we performed bulk RNA-seq of the BRAF V600E mouse melanoma cell line 5555 and its vemurafenib-resistant derivative 5555R, generated in our laboratory by prolonged exposure to vemurafenib. Compared with parental cells, resistant cells showed enrichment of inflammatory and interferon-response, cytoskeletal/actomyosin, mechano-sensing and epithelial-to-mesenchymal transition programmes.

These data accompany a study showing that BRAFi-resistant melanoma cells reactivate actomyosin contractility and adopt an altered secretory phenotype with increased M-CSF, which promotes differentiation of monocytes into immunosuppressive tumour-associated macrophages.
