ChIL-seq profiling of H3.3 and H3mm15 chromatin incorporation after histone chaperone depletion and amino acid substitution in NIH3T3 cells
Listed in NCBI GEO
H3mm15 is an H3.3-related histone variant that differs from H3.3 at three amino acid positions and exhibits distinct chromatin incorporation properties.
Description
To investigate the molecular mechanisms governing H3mm15 deposition and genomic localization, we performed Chromatin Integration Labeling followed by sequencing (ChIL-seq) in NIH3T3 cells expressing EGFP-tagged H3.3, H3mm15, or H3mm15 single-reversion mutants. The dataset consists of two complementary experiments.
First, H3.3-EGFP- and H3mm15-EGFP-expressing cells were treated with siRNAs targeting the histone chaperones Daxx or Hira and compared with cells treated with a non-targeting control siRNA. The genomic incorporation of the EGFP-tagged histones was profiled by GFP-ChIL-seq. Second, the genomic distributions of H3mm15-EGFP and three single-reversion mutants, Q100L, V112I, and C128R, were compared with that of H3.3-EGFP.

These data enable evaluation of the dependence of H3.3 and H3mm15 chromatin incorporation on the HIRA and DAXX histone chaperone pathways and assessment of how the H3mm15-specific amino acid residues influence its chromatin incorporation and genome-wide distribution.
