Effect of the Acr1 protein of Mtb on bone marrow cells cultured in the presence of GM-CSF and IL-4 and generation of MDSCs

Mycobacterium tuberculosis (Mtb) effectively suppresses host immunity to ensure its survival.
Description
We have earlier shown that the Acr1 protein of Mtb can inhibit the differentiation of dendritic cells (DCs). Consequently, in the current study, we examined the role of Acr1 in mitigating autoimmunity.
Initially, we observed that Acr1 skews the differentiation of DCs to functionally competent myeloid-derived suppressor cells (MDSCAcr1). MDSCAcr1 chiefly secretes immunosuppressive molecules, expands the pool of regulatory T cells (TregAcr1), and attenuates inflammatory responses. Further, MDSCAcr1 suppresses the differentiation and function of Th17 cells and the proliferation of syngeneic and alloreactive CD4 T cells and CD8 T cells.

These in vitro results were substantiated by injecting Acr1 in a murine experimental autoimmune encephalomyelitis (EAE) model. Interestingly, expansion in the MDSCs population with a concurrent increase in MOG-specific Tregs and a decline in Th17 cells was observed. Furthermore, alleviation of the clinical symptoms in EAE animals was observed.
These results were further validated in the prophylactic model of EAE. Consequently, Acr1 demonstrates a prophylactic effect by averting the onset of the disease. Mechanistically, Acr1 activates Tregs and MDSCs via the TLR-4 pathway, implicating innate immune recognition in Mtb-induced suppression.
The results indicate a potential role of Acr1 against autoimmune diseases.
