Type 1 lymphocytes accumulate in the thalamus and produce interferon-gamma to restrict seizure susceptibility after brain injury.

Listed in NCBI GEO

Interferons induce the transcription of a canonical set of interferon-stimulated genes upon binding to their receptor; however, whether these same genes are upregulated within CNS cell types in vivo remained unclear.

Description

To determine what the effects of IFNγ signaling in microglia in vivo, we injected IFNγ (or PBS vehicle control) into the brain of mice and isolated microglia for bulk RNA sequencing 22 hours after injection.

Differential gene expression analysis was then performed to determine which genes were upregulated or downregulated following IFNγ administration. Differentially upregulated genes were then used to construct a microglial IFNγ gene signature that was applied to our other datasets to create a score for IFNγ-responiveness.
