Thiolated Recombinant Human Catalase for Treatment of Refractory Dry Eye Disease, from Conceptual Design to First-in-human Study

To investigate the similarities and differences in the therapeutic mechanisms between hCAT-SH eye drops and cyclosporine eye drops in a benzalkonium chloride (BAC)-induced mouse model of dry eye disease (DED), we performed this transcriptomic sequencing study.
Description
Total RNA was extracted from eyeball tissues of Balb/c mice subjected to the following conditions: (1) BAC-induced DED model, (2) DED model treated with hCAT-SH eye drops, and (3) DED model treated with cyclosporine eye drops.
Each treatment was administered for seven days. A total of 12 samples (4 biological replicates per group) were subjected to strand-specific RNA sequencing on an Illumina NovaSeq™ 6000 platform. Analysis of the data identified 351 and 373 differentially regulated genes in the hCAT-SH and cyclosporine treatment groups, respectively.

A key finding is that exogenous hCAT-SH treatment did not significantly activate endogenous antioxidant regulatory pathways. Instead, it markedly suppressed genes involved in antigen processing and presentation as well as chemokine signaling pathways. In contrast, cyclosporine treatment activated the antigen processing and presentation pathway.
