RNA-seq Profiling of Activated Splenic T Cells from Control and Diet-Induced Obese Mice

Listed in NCBI GEO

Obesity is associated with impaired T cell function and may constrain the efficacy of adoptive cell therapies such as CAR-T cell therapy.

Description

To characterize obesity-induced transcriptional alterations in T cells prior to CAR transduction, we performed bulk RNA-seq on splenic T cells isolated from control (CON) or diet-induced obese (DIO) C57BL/6 female mice and activated ex vivo for 48 hours with anti-CD3/CD28 agonistic antibodies.

Transcriptomic profiling revealed enrichment of gene sets associated with negative regulation of T cell activation, effector function, and TGF-beta signaling in T cells from obese mice, consistent with an obesity-imprinted suppressive transcriptomic state in activated T cells.
