Super tRNA-Tumor suppressor gene-Manual

Based on the COSMIC database, we compiled all reported nonsense mutation types for the four tumour suppressor genes TP53, RB1, PTEN, and APC, and established corresponding super tRNA-manuals for each of these genes. We predicted the missense mutation effects for the saturated mutagenesis of each nonsense mutation site and generated a heatmap corresponding to each nonsense mutation.

The heat map corresponds to the predictive impact of each variant ranging from -1 (Red, Pathogenic) to 1 (Blue, Benign), with the lowest score being the most deleterious. Scores were obtained from nine algorithms, including PredictSNP, MAPP, PhD-SNP, PolyPhen-1, PolyPhen-2, SIFT, SNAP, PANTHER and Alphamissense. Based on this analysis, we provide tailored sup-tRNA selection recommendations for therapeutic targeting of each nonsense mutation. </p>
