Transcriptomic profiling of gastrointestinal cancer cells treated with the Pt(II)-Schiff base complex [PtCl(L1H)]Cl

Listed in NCBI GEO

Gastric cancer (GC) remains a major cause of cancer-related mortality and novel platinum-based agents with improved stability, solubility, and anticancer activity are still needed.

Description

In this study, we investigated the transcriptomic effects of the Pt(II)-Schiff base complex [PtCl(L1H)]Cl in gastrointestinal cancer models. MKN28, MKN45, and PANC1 cells were treated with [PtCl(L1H)]Cl for 24 h and subjected to RNA sequencing (RNA-seq).

Differential expression and pathway enrichment analyses revealed transcriptional programs associated with DNA damage response, apoptosis, oxidative stress, and epithelial-to-mesenchymal transition (EMT). Functional assays further demonstrated that [PtCl(L1H)]Cl impaired migration, invasion, and cancer stem-like cell properties. These data provide transcriptomic evidence supporting the antitumor and anti-CSC activity of [PtCl(L1H)]Cl and highlight ligand stabilization as a promising strategy for the development of next-generation platinum-based anticancer agents.
