RNA-seq analysis of transcriptomic changes in TRIP13-deficient MOLM-13 leukemia cells

We performed RNA-seq on MOLM-13 acute myeloid leukemia cells with TRIP13 knockdown to investigate the transcriptomic changes associated with this cancer-testis gene.

Description

TRIP13 is essential for AML progression but dispensable for normal hematopoiesis. Its activation reprograms cellular metabolism from glycolysis to oxidative phosphorylation, enhances fatty acid oxidation, and confers drug resistance.

Mechanistically, TRIP13 localizes to mitochondria, interacts with apoptosis-inducing factor (AIF), and reduces apoptotic priming. Disruption of the TRIP13-AIF interaction suppresses OXPHOS and reverses drug resistance.
