IL1A promotes inflammation while safeguarding epithelial basal cell identity and barrier function
Listed in NCBI GEO
The ability of barrier epithelia to sense physiological changes and initiate local inflammation is essential, as this process facilitates subsequent progenitor-mediated epithelial repair.
Description
Herein, we combine analysis of single-cell transcriptional RNA-sequencing from the human adult esophageal epithelium with spatially resolved characterization of epithelial cell populations. Using NGFR and P73, we define a layer of human basal progenitor cells spanning both papillary and interpapillary epithelial regions.
In contrast, cycling basal progenitor cells are enriched within the papillary epithelium and a subset of papillary basal progenitor cells are marked by the differentiation marker YAP, revealing spatially organized human progenitor cell heterogeneity. When challenged, NGFR-positive basal cells outcompete NGFR-negative suprabasal cells in primary organoid-forming capacity and suppress NOTCH signaling to maintain their basal identity.

Examining changes in progenitor cell states associated with esophageal regeneration and inflammation, we identify upregulation of IL1A in primary human organoids and find an intraepithelial apical-to-basal IL1A signaling loop active in human eosinophilic esophagitis patients. Mechanistically, IL1A activates downstream NF-kB, inducing epithelial expression of secreted mediators that recruit inflammatory immune cells, thereby amplifying and extending the inflammatory response.
Notably, IL1A also expands the basal progenitor cell pool and enhances epithelial barrier integrity. Together, these findings demonstrate how inflammatory signals can be harnessed to promote regeneration of the human esophageal epithelium.
