Neurotropic virus-induced astrocyte remodeling links collagen-rich extracellular matrix to the LINC00460–miR-4443 regulatory axis [nCounter Flex]

Astrocytes are critical regulators of central nervous system homeostasis and responses to viral infection, yet how the extracellular environment shapes astrocyte susceptibility to neurotropic viruses remains poorly understood.
Description
Here, we investigated the temporal and molecular consequences of human coronavirus 229E (HCoV-229E) infection in primary human astrocytes using integrated virological, ultrastructural, transcriptomic, proteomic, small-RNA and quantitative imaging approaches.
HCoV-229E established productive infection accompanied by extensive remodeling of intracellular membranes and the extracellular matrix (ECM). Multi-omic analyses identified collagen-rich ECM as a prominent and coordinated target of infection, with progressive depletion of collagen I. Collagen loss was reproduced following infection with the phylogenetically distinct HCoV-OC43 and the neurotropic Zika virus Puerto Rico strain (ZIKV-PR).

Importantly, increasing collagen abundance significantly reduced infection by HCoV-229E, HCoV-OC43 and ZIKV-PR, revealing a reciprocal relationship between collagen homeostasis and neurotropic viral susceptibility. Small-RNA profiling further identified an infection-associated LINC00460–miR-4443 regulatory axis, previously implicated primarily in cancer-associated cellular remodeling. Increasing miR-4443 altered astrocyte morphology, collagen I abundance and activation markers.
Together, these findings identify the collagen-rich ECM as an active component of astrocyte antiviral defense and uncover a previously unrecognized regulatory connection between neurotropic coronavirus infection, astrocyte remodeling and the LINC00460–miR-4443 axis.
