Small non-coding RNA profiling in the hippocampus of mice with postoperative cognitive dysfunction

Postoperative cognitive dysfunction (POCD) commonly affects elderly surgical patients and produces declines in memory, attention, and executive function that substantially reduce quality of life.
Description
The precise pathogenesis of POCD remains unclear. Recent studies implicate trauma-induced epigenetic mechanisms in its development.
Conventional sequencing methods are hindered by reverse-transcription arrest at modified RNAs, which often causes many small noncoding RNAs (sncRNAs) with key regulatory roles to be missed. Here, we apply PANDORA-seq to generate a comprehensive, unbiased profile of highly modified sncRNA expression in the hippocampus of a mouse model of POCD. The dataset documents dynamic changes in hippocampal sncRNA classes—miRNAs, tsRNAs, rsRNAs, and piRNAs—between control and surgical stress conditions.

To demonstrate the dataset’s reliability and exploratory value, we performed initial downstream analyses and technical validations using tsRNAs that exhibit pronounced dynamic restructuring as exemplars. Functional enrichment of predicted target genes for these differentially expressed tsRNAs linked them to key pathological processes, including dendritic spine morphogenesis, synaptic transmission, and necroptosis. Parallel qRT-PCR validation and structural analysis of the parental tRNAs confirmed the dataset’s high accuracy.
Overall, this resource provides a robust foundation for detailed investigation of the neuro-epigenetic mechanisms that drive POCD pathogenesis.
