The hsa_circ_0031607 Restricts Dengue Virus Replication in Association with PARP1 and Ferroptosis
Listed in NCBI GEO
Dengue virus (DENV) infection remains a global health threat without specific antiviral therapy.
Description
Although circular RNAs (circRNAs) are increasingly recognized in antiviral immunity, their roles during DENV infection are largely undefined. Here, we performed transcriptome sequencing of PBMCs from dengue patients stratified by disease phase and severity, identifying differentially expressed circRNAs across severity groups and clinical phases.
Cross-stage intersection identified 15 circRNAs dysregulated across all phases, enriched in ferroptosis, glutathione metabolism, and endocytosis. Among these, hsa_circ_0031607 (circSNX6) was the only one associated with all three pathways and was significantly upregulated in both dengue patients and DENV2-infected HUVECs. circSNX6 overexpression suppressed DENV2 replication, whereas its knockdown enhanced viral propagation. circSNX6 localized to the cytoplasm and physically interacted with PARP1.

DENV infection was accompanied by partial redistribution of PARP1 from the nucleus to the cytoplasm. circSNX6 suppressed PARP1 expression, and its knockdown was accompanied by altered global PARylation, including increased NRF2 PARylation and changes in TFR1 and GPX4 expression. In mice, pharmacological PARP1/2 inhibition with olaparib attenuated DENV pathology, accompanied by ferroptosis suppression. Pharmacological ferroptosis inhibition reduced DENV2 replication, whereas ferroptosis induction enhanced it, suggesting that the cellular ferroptotic state correlates with DENV2 replication.
Collectively, these findings provide a circRNA expression profile across dengue disease progression and suggest that circSNX6 restricts DENV2 replication, an effect associated with PARP1 interaction and ferroptosis modulation.
