PLC-IP3 signaling regulate sensory hair cell formation in zebrafish neuromast

Listed in NCBI GEO

Hearing loss typically arises from irreversible damage to sensory hair cells (HCs) in the inner ear.

Description

Currently, restoring the number and function of HCs in mammals remains a significant challenge, hindered by an inadequate understanding of the molecular mechanisms governing the differentiation of sensory HCs. Therefore, identifying novel regulators that promote supporting cell (SC) differentiation and HC formation is crucial in the field.

Phosphoinositide-specific phospholipase C (PLC) converts extracellular cues into IP3 and DAG to activate Ca²⁺/PKC signaling that drives cell-state transitions in proliferation, differentiation, and repair. However, the role of PLC in HCs differentiation has remained poorly defined in the context of HCs lineage progression and maturation. Here, we found that inhibiting PLC signaling reduces HC output from the progenitor pool, while activating the pathway enhances HC formation.

Among PLC isoforms, plcd4a is selectively enriched in HCs and is required for HCs formation. Mechanistically, we demonstrate that these processes are mediated through the Notch/Wnt-PLC-IP3 signaling axis. These findings identify PLC–IP3 signaling as a regulatory node linking HC formation with maturation.
