Multiomic and integrative analysis of human and murine AML data reveal a stem-like profile of Phf19-depleted cells
Listed in NCBI GEO
Acute myeloid leukemias (AMLs) are a type of cancer where undifferentiated myeloid cells abnormally proliferate and accumulate in the organism.
Description
Such disease may originate by the expression of oncogenic chimeric proteins, like MLL-AF9. Many works molecularly characterized the final stage of the leukemias, but fewer it’s known about the initiation process and the impact that cell-intrinsic factors (like epigenetics) may have on its development.
Indeed, studies focusing on early leukemogenic stages in epigenetically altered hematopoietic cells are lacking. Recently, we characterized the role of a Polycomb-associated protein, called Phf19 in healthy hematopoietic cells, whose depletion (Phf19KO) compromises the hematopoietic stem cells (HSCs) functioning, like cell state regulation and maintenance. Here, we expressed the MLL-AF9 oncogene in Phf19KO hematopoietic cells and generated multiomic data using bulk and single cell NGS technologies.

Overall, we observed the acquisition of stem-like properties at multiple levels in MLL-AF9_Phf19KO cells. Such results were extensively compared and integrated with the AML patient’s ones. This analysis confirmed the molecular and functional results we produced.
Finally, we identified a novel AML prognosis-predicting signature that highlighted the presence of few biomarkers not been described in AMLs yet.
