The role of TPM1 isoform1.1 and 1.6 on global splicing events of mice heart

Listed in NCBI GEO

This study aimed to investigate the role of alternative splicing of TPM1 exon 9a in mice heart.

Description

We utilized adeno-associated virus serotype 9 (AAV9) vectors under the control of the cardiac troponin T (cTnT) promoter to deliver two major cardiac TPM1 isoforms—TPM1a (variant 1.1, containing 9a) and TPM1b (variant 1.6, lacking exon 9a)—into cardiomyocytes of mouse. Each mouse received 3×10¹¹ viral genomes of the respective AAV9 construct.

At the experimental endpoint, bulk RNA sequencing was performed on cardiac tissue to assess global splicing changes. Our analysis revealed that exogenous expression of either TPM1 isoforms did not significantly alter global splicing patterns.
