A Cost-Effective Nuclei Isolation Protocol for Single Nucleus RNA-Sequencing of Mouse HFpEF Hearts

Single cell or single nuclei RNA-sequencing (snRNAseq) enables high-resolution profiling of cellular states, but sample preparation is often costly and complicated by debris formation and cell clumping.

Description

We describe a cost-effective approach using commercially available buffers and standard laboratory equipment to isolate high-quality nuclei from diseased cardiac tissue for reproducible downstream snRNAseq analysis. In addition, we include a previously unpublished heart failure with preserved ejection fraction (HFpEF) vs Chow dataset generated using this protocol.
