With the intensification of population aging, its incidence rate keeps rising. Microglia play a significant role in regulating neuronal functions.

Exosomes, as carriers of intercellular communication, can mediate intercellular information transmission and signal regulation. This study aims to explore the molecular mechanism by which Acanthopanax senticosus extract (AS) interferes with the PD process by regulating microglial exosome communication. The PD-related inflammation model was established by inducing microglial cell activation with lipopolysaccharide (LPS).

Exosomes derived from microglia were isolated by ultracentrifugation. The exosomes were co-cultured with PD cells to detect the expression levels of α-synuclein(α-syn) and Tyrosine Hydroxylase(TH) in PD cells. Transcriptome sequencing using exosomes revealed the mechanism by which AS treats PD by regulating the LncRNA-miRNA-mRNA network in exosomes, and the expression of target genes differentially expressed by exosomes was verified by the qRT-PCR method.

This study explores at the molecular level the mechanism by which Acanthopanax senticosus extract regulates microglial exosomes in communication therapy for Parkinson's disease, providing new research directions and theoretical basis for finding therapeutic targets for Parkinson's disease.
