Transcriptomic profiling of osteosarcoma patients with good and poor response to MAP regimen

Purpose: The overall objective of this study is to identify the differentially expressed genes and transcriptomic features associated with chemotherapy response in osteosarcoma patients treated with the MAP regimen (Methotrexate, Doxorubicin, and Cisplatin).

Methods

High-throughput transcriptome sequencing (RNA-seq) was performed on 9 osteosarcoma patient tumor tissue samples. Based on histological assessment, samples were categorized into 4 'good responders' (≥90 tumor necrosis) and 5 'poor responders' (≤60 tumor necrosis) to evaluate transcriptomic profiles corresponding to therapeutic responsiveness.Results: Using an optimized bioinformatics analysis pipeline, we identified key differentially expressed genes (DEGs) and enriched signaling pathways distinguishing good from poor responders.

These findings provide valuable insights into the transcriptomic landscape of chemoresistance and potential biomarkers for osteosarcoma therapeutic strategy.
