Peripheral double-negative T cells are enriched in the ovary and uterus and decline in mouse models of autoimmunity and IFN-γ-driven inflammation

Conventional NK1.1⁻ αβ DNTs are enriched in the mouse ovary and uterus and show a regulatory-like, CD8-distinct transcriptional program (low Cd8a/Cd8b1, Il7r, and cytotoxic genes; high Pdcd1, Lag3, Tox, Il10).

Description

Ex vivo, splenic DNTs have low effector output and suppress CD8⁺ T-cell proliferation mainly via contact-dependent mechanisms. In vivo, reproductive-tissue DNTs show tissue-restricted behavior, decline in IFN-γ-driven and ZP3-induced ovarian inflammation, and the DNT:CD8⁺ T-cell balance shifts toward activated CD8⁺ T cells.

Adoptive transfer of WT DNTs improves pregnancy frequency in IFN-γ-mediated infertility.
