Single-cell transcriptomic and organoid multi-omic profiling of NF1-regulated malignant progression in HPV-associated cervical epithelial lesions

This study was designed to investigate the cellular and molecular mechanisms underlying malignant progression of HPV-associated cervical epithelial lesions, with a particular focus on the role of NF1 in restraining oncogenic RAS signaling and malignant transformation.
Description
The study integrates single-cell RNA sequencing of primary human cervical tissues with ATAC-seq and bulk RNA-seq profiling of NF1-deficient and control human cervical precancerous organoids.
For single-cell RNA sequencing, fresh cervical tissues were collected from individuals with normal cervix (CON, n=10), low-grade or high-grade squamous intraepithelial lesions (SIL, n=19), and cervical squamous cell carcinoma (CC, n=23), for a total of 52 primary cervical tissue samples. For ATAC-seq, three NF1-knockout cervical precancerous organoid samples (sgNF1, n=3) and three non-targeting sgRNA control samples (sgNC, n=3) were analyzed.

For bulk RNA-seq, three NF1-knockout cervical precancerous organoid samples and three corresponding control samples were analyzed. Together, these datasets enable integrated analysis of epithelial state transitions, NF1-regulated transcriptional programs, RAS-associated signaling, and chromatin accessibility changes during cervical malignant progression.
