VAT CD4+ T cell responses in the absence of either tTreg or pTreg cells

Foxp3⁺ regulatory T (Treg) cells are important regulators in maintenance of visceral adipose tissue (VAT) homeostasis.

Description

The relative contributions of thymus-derived (tTreg) and peripherally induced (pTreg) subsets to the VAT Treg pool have remained unclear due to the absence of definitive lineage markers. To dissect their roles, we utilized complementary mouse models selectively deficient in either tTreg (ΔtTreg) or pTreg (ΔpTreg) cell development.

CD4⁺ T cells were FACS-purified from VAT of these mice and analyzed using single-cell RNA sequencing (scRNA-seq) and single-cell T cell receptor sequencing (scTCR-seq) via the 10x Genomics. This dataset provides a high-resolution view of VAT CD4⁺ T cell subsets and TCR clonotypes, enabling functional insights into how pTreg and tTreg cells differentially contribute to adipose tissue homeostasis and inflammation.
