Single-cell RNA-seq reveals the keratinocyte heterogeneity and increased interfollicular basal keratinocytes in wound-induced hair follicle neogenesis

Listed in NCBI GEO

Scarring alopecia is an unresolved challenge characterized by scar formation following skin injury without hair follicle (HF) regeneration.

Description

Wound-induced hair follicle neogenesis (WIHN) is a model in which HFs regenerate after skin wounding. This study aimed to explore the mechanisms underlying WIHN, hinting for the new therapeutic strategies for scarring alopecia.

Our results indicated that keratinocytes were categorized into 8 subpopulations, with a notable increase in the proportion of INFU basal KCs in tissue with WIHN, which was validated by IF staining. Pseudotime analysis revealed that the marker gene Fst of INFU basal KC was activated earlier in tissue with WIHN. Fibroblasts were classified into 5 subpopulations, with an increase in the proportions of secretory reticular fibroblasts.

Cell-cell communication analysis showed that interactions between INFU basal KC and secretory reticular fibroblasts were significantly enhanced. Enrichment analysis further demonstrated that extracellular matrix (ECM)-receptor interactions were significantly enriched in ligand-receptor interactions. Masson staining exhibited decreased collagen content in tissue with WIHN.These findings suggest that the importance of INFU basal KCs in WIHN and that the ECM reconstruction is crucial for WIHN.

Our study contributes to a better understanding of the underlying mechanisms of WIHN.
