DMSO-induced human embryo model reveals V-ATPase requirement in cavitation

This study focuses on the key regulatory mechanisms of lysosomal acidification in early human embryonic development.

Description

By employing single-cell RNA sequencing technology (Smart-seq2), we conducted a systematic analysis of human morulae derived from three pronuclei, embryos arrested in development after treatment with 1 nM Concanamycin A, and control blastocysts. Through sequence alignment and quantitative analysis based on the human reference genome, the study successfully revealed the significant biological role of lysosomal acidification in the differentiation process of the trophectoderm (TE).

This finding provides a new theoretical perspective for a deeper understanding of the molecular mechanisms underlying embryonic developmental arrest.
