An advanced scRNA-seq-compatible lentiviral barcoding system resolves treatment-associated clonal selection and transcriptional states in HNSCC cells

Listed in NCBI GEO

Cancer therapy response is shaped by pre-existing heterogeneity and treatment-associated selection within tumor cell populations.

Description

In this study, we applied a single-cell RNA-seq-compatible lentiviral BC24 barcode system to FaDu hypopharyngeal squamous cell carcinoma cells to link clonal identity with transcriptional cell state during cytotoxic treatment response. Barcoded FaDu cells were profiled by 10x Genomics single-cell RNA sequencing together with targeted recovery of expressed BC24 lineage barcodes from amplified cDNA.

The dataset enables analysis of untreated and treatment-selected FaDu cells, including cisplatin- and irradiation-associated transcriptional remodeling, barcode-defined clonal enrichment, and the relationship between therapy-associated clonal selection and pre-existing epithelial cell states.
